چکیده

Interstitial ̄uid pressure (IFP) is one of the main obstacles for macromolecular agents uptake and distribution in solid tumors. It has been demonstrated to reduce e€ectiveness of different macromolecular agents used in in vivo anti-tumor therapies, which on the other hand showed very good anti-tumor properties in in vitro conditions [L.T. Baxter, R.K. Jain, Microvascular Research 37 (1989) 77±104]. With an appropriate model we demonstrated the correlation between di€erent physiological properties of solid tumor and IFP. The model which we present showed high correlation with results from literature and thus represents a good simulation of physiological processes that govern ̄uid dynamics in solid tumors. One of the potential uses of presented model is drafting of future experiments which would lead to more e€ective chemoor immuno-therapy of solid tumors. This model could also serve as an aid to the interpretation of di€erent experimental results concerning IFP. Ó 2000 Published by Elsevier Science B.V. All rights reserved.

Numerical Modeling of Fluid Flow in Solid Tumors

A mathematical model of interstitial fluid flow is developed, based on the application of the governing equations for fluid flow, i.e., the conservation laws for mass and momentum, to physiological systems containing solid tumors. The discretized form of the governing equations, with appropriate boundary conditions, is developed for a predefined tumor geometry. The interstitial fluid pressure a...

Mathematical Model of the Effect of Interstitial Fluid Pressure on Angiogenic Behavior in Solid Tumors

We present a mathematical model for the concentrations of proangiogenic and antiangiogenic growth factors, and their resulting balance/imbalance, in host and tumor tissue. In addition to production, diffusion, and degradation of these angiogenic growth factors (AGFs), we include interstitial convection to study the locally destabilizing effects of interstitial fluid pressure (IFP) on the activi...

Numerical Modeling of Interstitial Fluid Flow Coupled with Blood Flow through a Remodeled Solid Tumor Microvascular Network

Modeling of interstitial fluid flow involves processes such as fluid diffusion, convective transport in extracellular matrix, and extravasation from blood vessels. To date, majority of microvascular flow modeling has been done at different levels and scales mostly on simple tumor shapes with their capillaries. However, with our proposed numerical model, more complex and realistic tumor shapes a...

Time-dependent behavior of interstitial fluid pressure in solid tumors: implications for drug delivery.

Elevated interstitial fluid pressure (IFP) may constitute a significant physiological barrier to drug delivery in solid tumors. Strategies for overcoming this barrier have not been developed to date. To identify and characterize various mechanisms regulating IFP and to develop strategies for overcoming the IFP barrier, we modeled the tumor as a poroelastic solid. We used this model to simulate ...

Oncotic pressure in solid tumors is elevated.

Oncotic and hydrostatic pressure differences control the movement of fluid and large molecules across the microvascular wall of normal and tumor tissues. Recent studies have shown that the interstitial fluid pressure in tumors is elevated and is approximately equal to the microvascular pressure. Whereas oncotic pressure in blood plasma of various species is known, no data are available on the o...

Effect of Fluid Friction on Interstitial Fluid Flow Coupled with Blood Flow through Solid Tumor Microvascular Network

A solid tumor is investigated as porous media for fluid flow simulation. Most of the studies use Darcy model for porous media. In Darcy model, the fluid friction is neglected and a few simplified assumptions are implemented. In this study, the effect of these assumptions is studied by considering Brinkman model. A multiscale mathematical method which calculates fluid flow to a solid tumor is us...
